Vitamin D, specifically cholecalciferol (Vitamin D3), is a critical nutrient involved in calcium and phosphorus metabolism across all livestock species. It can be supplied through endogenous synthesis in the skin via ultraviolet (UV) light exposure or, more commonly in modern production systems, through dietary supplementation. The use of 25-hydroxycholecalciferol (25-OH D3) for diet supplementation, which is the predominant circulating metabolite of Vitamin D3 required for physiological functions, has become standard practice in swine, poultry, and bovine nutrition due to its superior bioavailability and metabolic efficiency.

However, not all dietary sources of 25-OH D3 are equivalent in purity, quality, potency, or stability. Several key factors must be considered when evaluating their inclusion in feed formulations, including:

Cholecalciferol (Vitamin D3) is biologically inactive and requires a two-step enzymatic conversion to yield its active form, 1,25-dihydroxycholecalciferol (1,25(OH)2D3). This metabolite functions as a steroid hormone and is tightly regulated within the animal’s endocrine system. It exerts its physiological effects by modulating the expression of numerous genes, including those involved in calcium and phosphorus homeostasis and absorption.

Due to the hormone’s narrow and regulated range of physiological necessity, the exogenous inclusion of 1,25(OH)2D3 through the diet can override endogenous control mechanisms, leading to negative effects. The most observed response to elevated levels is impaired growth performance resulting from excessive calcium absorption and increased risk for hypercalcemia. This condition is extremely detrimental, contributing to kidney and liver damage, and substantial increases in mortality in severe cases.

To ensure animal safety and optimal physiological function, it is critical that sources of dietary 25-hydroxycholecalciferol (25-OH D3) are free of contaminating levels of 1,25(OH)2D3. According to FDA regulations (CFR Vol. 83 No. 191) for supplementation in approved species, the concentration of 1,25(OH)2D3 must not exceed 1 part per billion (ppb) in a 25-OH D3 product that must have at least 94% purity for 25-OH D3.

Reliability

Utilizing an accredited laboratory is a critical strategy for routinely verifying product concentration, quality and safety to ensure traceability throughout the supply chain. While quantifying the concentration of 25-hydroxycholecalciferol (25-OH D3) is a primary analytical target, it should not be the sole criterion for evaluation. Comprehensive product assessment requires the measurement of additional biochemical markers that validate the manufacturing processes and reflect both the safety and bioactivity of the product.

Foremost among these is 1,25-dihydroxycholecalciferol (1,25(OH)2D3), the active hormonal form of Vitamin D3, which must be rigorously assayed due to its potent physiological effects and tight endogenous regulation. Presence of 1,25(OH)2D3 above threshold levels may indicate metabolic contamination and pose safety risks.
